Mechanism of Action: Tesofensine functions by inhibiting the reuptake of three key neurotransmitters: serotonin, noradrenaline, and dopamine. This inhibition leads to increased concentrations of these neurotransmitters in the synaptic cleft, enhancing neurotransmission and resulting in appetite suppression and potential weight loss.
Clinical Development: Initially, tesofensine was explored for treating Alzheimer's and Parkinson's diseases. However, due to limited efficacy in these areas, its development shifted focus to obesity treatment. Clinical trials have demonstrated significant weight loss in obese patients, with some studies reporting an average loss of 12.8 kg over six months at a 1 mg daily dose.
